The d-Amino Acid Oxidase, Uricase, and Choline Oxidase in Two Transplanted Rat Tumors and in Isolated Nuclei of Tumor Cells*
نویسنده
چکیده
In order to determine whether or not changes in enzymatic activity specifically accompany neoplasia, and to what extent such changes may occur, many enzymes have been investigated by Greenstein (2) in both normal and neoplastic tissues. Shack (3) has reported that d-amino acid oxidase is low in rat hepatoma 31, and suggested that the lowering of the enzyme may be owing to a deficiency in protein component as well as in the flavin prosthetic group of the enzyme, but no attempt has been made to determine the apoenzyme and coenzyme of d-amino acid oxidase separately. Furthermore, very little work has been done on the change in distribution of enzymes between the nuclei and cytoplasm of tumor tissue, owing to lack of availability of satisfactory preparations of tumor cell nuclei. In this work both the apoenzyme and coenzyme of d-amino acid oxidase have been studied in whole tumor tissue and in isolated nuclei of tumor cells. A study has been made, also, of the enzymes uricase and choline oxidase, two enzymes hitherto not investigated in tumor tissue.

A Study of d-Amino Acid Oxidase, Uricase, and Choline Oxidase in the Livers and in Isolated Liver Cell Nuclei of Rats Bearing Transplanted Tumors*
Since the report of Brahn (1) that very low catalase values are found in the livers of human beings dead of various forms of cancer, many papers have appeared on the effect of a growing tumor upon enzyme systems in liver tissue. Greenstein (4) has shown that in rats bearing transplanted hepatoma 31 or Jensen sarcoma, or in mice bearing various transplanted tumors, the activity of the liver cata...
متن کاملPhylogeny of urate oxidase producing bacteria: on the basis of gene sequences of 16S rRNA and uricase protein
Uricase or Urate oxidase (urate:oxygen oxidoreductase, EC 1.7.3.3), a peroxisomal enzyme which is found in many bacteria, catalyzes the oxidative opening of the purine ring of urate to yield allantoin, carbon dioxide, and hydrogen peroxide. In this study, the phylogeny of urate oxidase (uricase) producing bacteria was studied based on gene sequences of 16S rRNA and uricase protein. Repres...
متن کاملIntracellular distribution of uricase and catalase in livers of tumor-bearing mice and mice given injections of aminotriazole.
That the catalase activity of the liver of the tumor-bearing animal is significantly decreased has been established beyond question (6). The ac tivities of D-amino acid oxidase, argÃ1⁄4Ã-ase, sterase, and lipase are also diminished, although to a much lesser extent than catalase (6), while most other enzymes assayed have shown negligible changes in concentration. Uricase has not been studied so...
متن کاملComputational Approach for Rational Design of Fusion Uricase with PAS Sequences
Tumor lysis syndrome is a life-threatening condition for humans due to the lack of urate oxidase. In this study, several variants of PASylateduricasefrom the Aspergillus flavus species were analyzed computationally to find the appropriate fusions to solve short half-life and stability concerns. The Ab initio method was performed using Rosetta software to structurally characterize the PAS sequen...
متن کاملIdentification of Intracellular Sources Responsible for Endogenous Reactive Oxygen Species Formation
The endogenous reactive oxygen species ("ROS") formation is associated with many pathologic states such as inflammatory diseases, neurodegenerative diseases, brain and heart ischemic injuries, cancer, and aging. The purpose of this study was to investigate the endogenous sources for "ROS" formation in intact isolated rat hepatocytes, in particular, peroxisomal oxidases, monoamine oxidase, xanth...
